Novel Compound Reduces Thrombosis Risk without Increased Bleeding Factor XI Antisense Oligonucleotide for Prevention of Venous Thrombosis[ LBA-1 ]
The four new kits, the VisuLize[TM] Factor VIII Antigen ELISA Kit, the VisuLize[TM] Factor IX Antigen ELISA Kit, the VisuLize[TM] Factor XI Antigen ELISA Kit, and the VisuLize[TM] TAFI Antigen ELISA Kit, are intended to assess Factor VIII, Factor IX, Factor XI, and TAFI antigens in human plasma samples.
Importantly, factor VIII and factor IX deficiencies or severe factor XI deficiencies are associated with an increased risk of bleeding, whereas even drastic reductions of factor XII or rarely occurring high molecular weight kininogen and prekallikrein deficiencies do not cause excessive bleeding (4).
Most patients with congenital factor XI deficiency do not have any significant bleeding problems, although there are cases where such patients had significant hemorrhage after surgery or trauma.
Comparing 473 clot patients with 474 healthy participants matched for age and other characteristics, the researchers found that 92 of the patients but only 47 of the volunteers had factor XI concentrations exceeding 121 percent of normal.
Because Factor XI is involved in the propagation of clots, but not in their initiation, we hypothesized that reducing Factor XI activity would decrease the risk of deep vein thrombosis after knee replacement surgery without increasing the risk of bleeding.

Our testing revealed, however, that only the 1:10 dilution (1 volume of sample in 9 volumes of diluent) revealed a low factor XI activity, and the factor XI activity increased with dilution, such that a 1:40 dilution (1 volume sample in 39 volumes diluent) had normal factor XI activity.
